Just remember, it's really important not to hesitate.
Once you see an attractive girl, go for it right away! Once you're in, you're in.
The best way to deal with women after opening them with good pickup lines that I've found is to tease them a little bit, and treat them sort of like you would a bratty little sister.
Just be playful, and you'll get a good reaction from most girls.
